Building Proteins from Amino Acids
Think of amino acids as molecular Lego blocks that snap together to build every protein in your body. There are 20 naturally occurring amino acids, and they're found in all living things - which actually provides evidence for evolution since we all share this same basic toolkit.
Each amino acid has the same basic structure: an amino group (which is basic), a carboxyl group (which is acidic), and a unique side chain called the R group that makes each amino acid different. The carbon in the middle makes four bonds, connecting all these parts together.
When two amino acids join up, they form a peptide bond through a condensation reaction. The OH from one amino acid's carboxyl group combines with an H from another's amino group, releasing water and creating a dipeptide. Keep adding more amino acids and you'll get a polypeptide chain - the foundation of every protein.
Quick Test Tip: The Biuret test detects peptide bonds by turning purple/violet in alkaline conditions with a few drops of dilute copper sulfate!
Protein Structure - From Simple to Complex
Your body transforms simple amino acid chains into incredibly complex proteins through four levels of structure, each more sophisticated than the last. The primary structure is just the sequence of amino acids joined by peptide bonds - but this sequence determines everything that follows.
Secondary structure creates the first 3D shapes through hydrogen bonding. The slightly positive hydrogen from NH groups attracts the slightly negative oxygen from C=O groups, forming either alpha helices (coils) or beta pleated sheets. This gives proteins their basic architectural framework.
Tertiary structure is where proteins really come alive through three types of bonds: ionic bonding between charged amino acids, additional hydrogen bonding, and the strongest of all - disulfide bridges between sulfur atoms. This creates the final 3D shape that determines the protein's function.
Some proteins go even further with quaternary structure, combining multiple polypeptide chains with non-protein groups. Haemoglobin is a perfect example - four alpha helices each paired with an iron-containing haem group that actually carries your oxygen.
Remember: Fibrous proteins like collagen are long and structural (think tendons), while globular proteins like haemoglobin are compact and metabolic!
