Light Microscopes and Laser Scanning Confocal Microscopes
Light microscopes are your go-to tool for basic cell observation. They use a compound lens system with an objective lens and eyepiece lens that work together to magnify specimens up to ×1500 with a resolution of 200nm. Think of it as a sophisticated magnifying glass that lets you see living cells in action.
The beauty of light microscopy lies in its simplicity - you can observe living specimens without complex preparation. Samples are typically dehydrated with alcohol, embedded in wax blocks, then sliced with a microtome and stained with dyes like crystal violet to create colour contrast.
Laser scanning confocal microscopes take things up a notch by using focused laser light instead of regular illumination. A single laser spot moves across the specimen, and the microscope blocks out unwanted light from other areas. This creates incredibly sharp, clear images by examining only a very thin section at a time.

Electron Microscopes: TEM and SEM
Electron microscopes revolutionise what we can see by using electron beams instead of light, achieving magnifications up to ×500,000. However, there's a trade-off - all specimens must be dead due to the harsh preparation process required.
Transmission electron microscopes (TEM) work similarly to light microscopes but transmit electrons through ultra-thin specimens. The sample preparation is intense: specimens are killed through fixation, dehydrated with solvents, embedded in resin, sliced incredibly thin, then stained with heavy metals. The result? Incredible resolution of 0.5nm showing internal cellular structures in remarkable detail.
Scanning electron microscopes (SEM) bounce electrons off the specimen's surface rather than passing through it. This creates stunning three-dimensional images with a resolution of 3-10nm. Sample preparation involves similar steps to TEM, plus coating with heavy metals to enhance contrast.
Remember: Both electron microscopes typically produce black and white images, though these can be digitally coloured later for clarity.
The key difference between magnification and resolution is crucial here - magnification makes things bigger, whilst resolution determines how much fine detail you can actually see. Electron microscopes excel at both compared to light microscopes.
