Osnove človeške genetike
Človeška genetika preučuje, kako se lastnosti prenašajo iz staršev na otroke. To ni samo barva oči - gre tudi za nagnjenost k boleznim, telesno zgradbo in celo določene značajske lastnosti.
Kariotip je kot "fotografija" vseh tvojih kromosomov. Človek ima 46 kromosomov: 22 parov avtosomov in 1 par spolnih kromosomov (XX za dekleta, XY za fante). To zapišemo kot 46,XX ali 46,XY.
Rodovnik je družinsko drevo z znanstvenim pridihom. Uporabljamo ga za sledenje, kako se določena lastnost ali bolezen prenaša skozi generacije. Kvadrat predstavlja moškega, krog žensko, pobarvana figura pa označuje osebo z določeno lastnostjo.
💡 Pomni: Avtosomno dedovanje ni odvisno od spola, spolno vezano pa je - zato so nekatere bolezni pogostejše pri fantih!
Načini dedovanja pri človeku
Avtosomno dominantno dedovanje pomeni, da je za izražanje lastnosti dovolj že en dominanten alel. Huntingtonova bolezen je tak primer - lastnost se pojavi v vsaki generaciji in približno enako prizadene oba spola.
Avtosomno recesivno dedovanje je drugačno. Potrebuješ dva recesivna alela, da se lastnost izrazi. Cistična fibroza je klasičen primer - bolezen lahko "preskoči" generacijo, ker so starši lahko zdravi prenašalci.
Spolno vezano dedovanje je najpogosteje vezano na kromosom X. Ker imajo fantje samo en X kromosom, se pri njih izrazi vsak recesivni alel. Dekleta potrebujejo dva recesivna alela. Zato so hemofilija in daltonizem pogostejša pri fantih.
💡 Tip za test: Če v rodovniku bolezen močno prevladuje pri moških, pomisli na X-vezano recesivno dedovanje!
Genetske bolezni
Kromosomske bolezni nastanejo zaradi napačnega števila ali strukture kromosomov. Downov sindrom je posledica dodatnega kromosoma 21 (trisomija 21) - oseba ima torej 47 kromosomov namesto običajnih 46.
Monogenske bolezni so posledica mutacije v enem samem genu. Te se dedujejo po znanih vzorcih - dominantno, recesivno ali X-vezano. Cistična fibroza je avtosomno recesivna, Huntingtonova avtosomno dominantna.
Pri analizi rodovnikov iščeš vzorce. Avtosomno dominantne bolezni se pojavljajo v vsaki generaciji, recesivne lahko preskočijo, X-vezane pa močno prevladujejo pri moških.
💡 Ne zamenjaj: Downov sindrom ni genska bolezen, temveč kromosomska nepravilnost - gre za celoten dodaten kromosom!
Gensko inženirstvo
Gensko inženirstvo je kot "presajanje" genov iz enega organizma v drugega. Najbolj znan primer je proizvodnja človeškega inzulina v bakterijah - to je revolucioniralo zdravljenje sladkorne bolezni.
Postopek je presenetljivo logičen: iz človeške celice izoliraš gen za inzulin, ga vstaviš v bakterijski plazmid (majhen krožni DNK), nato pa ta rekombinantni plazmid vnesеš v bakterijo. Bakterije se hitro razmnožujejo in proizvajajo človeški inzulin.
Genetsko spremenjeni organizmi (GSO) so rezultat tega procesa. To ni samo inzulin - obstaja koruza, odporna na škodljivce, paradižnik z daljšo obstojnostjo in še veliko drugih.
💡 Ključni koraki: Izolacija gena → vstavljanje v vektor → transformacija → gojenje → čiščenje produkta
Genska terapija in etika
Genska terapija poskuša zdraviti bolezni z "popravljanjem" okvarjenih genov. To je kot zamenjava pokvarjenega dela v stroju, samo da gre za naš genetski material. Običajno se uporablja virus kot "prevozno sredstvo" za vnos zdravega gena.
Obstajata dva tipa: somatska genska terapija spreminja telesne celice (spremembe se ne dedujejo), zarodna genska terapija pa spolne celice ali zarodek (spremembe se dedujejo). Slednja je pri ljudeh prepovedana zaradi etičnih pomislekov.
Etična vprašanja so ključna. Ali je varno uživati GSO hrano? Kje je meja pri "izboljševanju" človeških lastnosti? Kdo bo imel dostop do dragih genskih terapij? To niso samo znanstvena, temveč tudi družbena vprašanja.
