Understanding Proteins for AQA A-Level Biology

Protein Structure and Building Blocks
Amino acids are the basic building blocks of all proteins - think of them as biological Lego bricks. There are 20 naturally occurring amino acids, and each one has an amino group, a carboxyl group, and a unique side chain (called the R group) that gives it special properties.
When amino acids link together, they form peptide bonds through a process that removes water molecules. You can test for these bonds using the biuret test - it turns purple/violet if proteins are present, or stays blue if they're not.
Proteins have four levels of structure that determine how they work. The primary structure is simply the sequence of amino acids strung together like beads on a necklace. The secondary structure forms when this chain coils into regular patterns like the alpha helix, held together by hydrogen bonds.
Quick Test Tip: Remember the biuret test colours - purple means protein present, blue means no protein!
The tertiary structure happens when the protein folds into its final 3D shape, held by various bonds between the R groups. Finally, quaternary structure occurs when multiple protein chains work together, like in haemoglobin which has four separate chains plus special haem groups for oxygen transport.
There are two main protein types: globular proteins (ball-shaped and water-soluble like enzymes) and fibrous proteins (long, thin molecules like collagen in your tendons).
