Exploring Electron Microscopy: A Comparison with Light Microscopes

Types of Microscopes and Their Capabilities
Electron microscopes revolutionize our view of the microscopic world by using electron beams with wavelengths under 1nm. The Transmission Electron Microscope (TEM) focuses electrons through the specimen with electromagnets, creating detailed 2D images. Denser parts absorb more electrons, appearing darker in the image, and TEMs can achieve an impressive magnification of 500,000x with resolution of 0.0002μm.
The Scanning Electron Microscope (SEM) works differently, sending electrons across the specimen's surface and collecting reflected electrons. This creates detailed 3D images at magnifications up to 100,000x with 0.002μm resolution. Specimens for electron microscopy must be stained with heavy metals like gold, which scatter electrons to create contrast, with color added artificially afterward.
Light microscopes remain valuable tools despite lower magnification (up to 2,000x). They're inexpensive, portable, simple to operate, and can examine living specimens without requiring a vacuum. The Laser Scanning Confocal Microscope represents an advanced light microscope that uses laser beams to make fluorescent dyes in the specimen emit light, creating exceptionally clear images by blocking out-of-focus light through a pinhole.
